Ac-Leu-Lys-Lys-Thr-Glu-Thr-Gln. The N-terminus is acetylated; the C-terminus is a free acid. The acetyl group is not cosmetic — it is the difference between this molecule and the plain heptapeptide, and it accounts for 42 of the 889 daltons.

Origin

Thymosin β4 is a 43-residue protein found across many tissues. TB-500 is the acetylated form of its actin-binding stretch, residues 17 to 23. It is made by solid-phase synthesis, not isolated from tissue, and it is a fragment: anyone offering “TB-500” at a molecular weight near 4,900 is selling the full protein under a fragment's name.

Solubility

Soluble in water and in aqueous buffers. The two lysines and the glutamate make it amphoteric; solubility and net charge both move with pH.

Storage and handling

  • Lyophilised powder: store at −20 °C, protected from light and moisture.
  • Bring the vial to room temperature before opening, so condensation does not form on the powder.
  • In solution: refrigerate, protect from light, and treat the working life as far shorter than that of the powder.
  • Avoid repeated freeze-thaw cycles; aliquot first if the solution will be drawn on more than once.
  • Label every reconstituted vial with compound, lot, concentration and date.
