Semax

Four residues borrowed from a hormone fragment, with a three-residue tail bolted on the end. The tail is the whole point of the molecule: it is what makes this a distinct compound rather than a piece of something else.

Met-Glu-His-Phe-Pro-Gly-Pro. Free at both termini. The first four residues are the ACTH(4–7) fragment; Pro-Gly-Pro is the synthetic C-terminal extension. The single methionine is the only sulphur in the molecule, and it is the residue most vulnerable to oxidation.

Origin

Semax was developed in Russia in the 1980s at the Institute of Molecular Genetics. Its parent fragment, ACTH(4–7), is a stretch of adrenocorticotropic hormone; the Pro-Gly-Pro tail was added deliberately and does not appear in the hormone. It is made entirely by synthesis.

Solubility

Soluble in water and in aqueous buffers. Neutral to slightly acidic conditions are gentler on the methionine than alkaline ones.

Storage and handling

  • Lyophilised powder: store at −20 °C, protected from light and moisture.
  • Methionine oxidises. Minimise headspace air, keep the vial closed, and do not leave solutions standing open.
  • In solution: refrigerate, protect from light, and treat the working life as far shorter than that of the powder.
  • Avoid repeated freeze-thaw cycles; aliquot first if the solution will be drawn on more than once.
  • Label every reconstituted vial with compound, lot, concentration and date.
