Structure
adenine – ribose – phosphate – phosphate – ribose – nicotinamide
Adenosine monophosphate and nicotinamide mononucleotide, joined through a pyrophosphate bridge. The oxidised form, NAD+, carries a positive charge on the nicotinamide ring; the reduced form, NADH, does not, and the two are distinguishable by ultraviolet absorbance at 340 nm. What is supplied here is the oxidised form.
Origin
NAD+ is a coenzyme present in every living cell, described in the early twentieth century and among the most studied molecules in biochemistry. Material of this grade is produced by fermentation or enzymatic synthesis and purified, rather than being extracted from tissue.
Solubility
Freely soluble in water — substantially more so than any peptide in this catalog. Solutions are acidic; the pyrophosphate bridge is the least stable part of the molecule and hydrolyses faster as pH and temperature rise. Alkaline conditions degrade it quickly.
Storage and handling
- Powder: store at −20 °C, protected from light and moisture. Hygroscopic — keep the vial closed and equalise to room temperature before opening.
- Avoid alkaline buffers. The pyrophosphate bond hydrolyses readily above neutral pH.
- In solution: refrigerate, protect from light, use promptly, and do not hold warm.
- Avoid repeated freeze-thaw cycles; aliquot first if the solution will be drawn on more than once.
- Label every reconstituted vial with compound, lot, concentration and date.
