Structure
trans-3-hexenoyl – [ human GRF 1–44 ] – NH2
The full 44-residue sequence of human growth hormone-releasing factor, amidated at the C-terminus as the natural hormone is, with a six-carbon unsaturated acyl group — trans-3-hexenoic acid — bonded to the N-terminal tyrosine. That acyl group is the entire difference between tesamorelin and the natural 44-mer, and it is what the name TH9507 identifies. One methionine accounts for the single sulphur.
Origin
Growth hormone-releasing factor is a hypothalamic peptide; its 1–44 amide form is the full-length natural species. Tesamorelin was developed by Theratechnologies. It is made by solid-phase synthesis and the acyl group added chemically — nothing here is extracted from tissue.
Solubility
Soluble in water and in aqueous buffers, though a 44-residue chain hydrates more slowly than a short peptide and rewards patience. The hexenoyl chain adds a hydrophobic head to an otherwise hydrophilic molecule, which is enough to make it more surface-active than its length alone would suggest — another reason not to shake it.
Storage and handling
- Lyophilised powder: store at −20 °C, protected from light and moisture.
- Direct the diluent down the wall of the vial. A stream driven into the cake will foam this one readily.
- In solution: refrigerate, protect from light, and treat the working life as far shorter than that of the powder.
- Avoid repeated freeze-thaw cycles; aliquot first if the solution will be drawn on more than once.
- Label every reconstituted vial with compound, lot, concentration and date.
